Haemostasis unit test repertoire and sample requirement

 

1. Thrombotic Investigations sample requirement:

For a full thrombophilia profile 4 green citrated and 1 serum gel samples are required. The serum gel sample should be sent to Immunology for Anti Cardiolipins.

For Factor V Leiden , Prothrombin (G20210A) and MTHFR only requests, a minimum one green citrated (recommended) or EDTA sample may be used as only the buffy coat is required. Two samples are preferred and separated buffy coats are also acceptable.

For Paediatric thrombophilia profile as much blood as possible is required. A minimum is 2 ml of blood with tubes correctly filled

 
2. Bleeding Investigations:

For a full bleeding profile a minimum of 20ml of fresh citrated blood is required. If a PFA, TEG and platelet nucleotides are also needed a further 10ml of blood should be sent.

Platelet Nucleotide samples should be received within two hours. If required alone 10ml citrated blood is required.

For a Factor assay one green citrated sample is required.

For a factor VIII inhibitor screen 3.0ml citrated blood is required.

For a factor VIII inhibitor assay 3.0ml citrated blood is required.

For a Low Molecular Weight Heparin Assay send one green citrated sample.

Platelet Aggregation Studies should not be more than 2 hours old. 20 ml citrated blood is required.

Samples for PFA should not be more than 4 hours old. 3ml citrated blood required.

ROTEM should not be more than 2 hours old. 3ml citrated blood required.

Samples for Thrombin Generation should not be more than 2 hours old. 3ml citrate blood contains Corn Trypsin Inhibitors required, Contact Haemostasis Lab for advice.

GTi Diagnostics HIT Elisa 5.0ml fresh serum should be tested while still fresh. Samples that cannot be tested immediately should be stored at 2 to 8°C for no longer than 48 hours or frozen. Samples frozen at -20°C or below remain in good condition for several years (2-3 years). Do not freeze/thaw.

For ADAMT13 Assay 10 ml of fresh citrated blood is required.

Euglobulin Clot Lysis Time should be placed on ice and received into the laboratory within 30 minutes of collection. A normal control should be collected at the same time. 20ml citrated blood required

Fibrin Plates Lysis should be placed on ice and received into the laboratory within 30 minutes of collection. A normal control should be collected at the same time. 20 ml citrated blood required.

Plasminogen Activator Inhibitor should be bleed into a BD Vacutainer CTAD tube and placed onto ice. Samples can be stored on ice for up to 7 hours in the collection tubes. If the sample is not tested immediately, it should be separated and frozen as soon as possible.
